Designates U.S. Highway 65 South from Pine Bluff, Arkansas, to the Arkansas-Louisiana border as the "Delta Rhythm & Bayous Highway"

Requires the Arkansas State Highway and Transportation Department to erect appropriate signs along the designated route, including signs in or near Dermott, Dumas, Gould, Lake Village, Pine Bluff, and Tillar

Authorizes the Department to accept gifts, grants, and donations from private, public, and nonprofit sources to fund acquisition, installation, maintenance, replacement, and reconstruction of the highway designation signs

Establishes legislative findings recognizing the southeast Arkansas Delta's rich musical heritage, influential musicians from the region, and the ecological and historical significance of Bayou Bartholomew and surrounding bayous
