Designates the portion of United States Highway 24 from Mile Marker 308 to Mile Marker 298 near Garden of the Gods through Colorado Springs and Manitou Springs as the "Buffalo Soldiers Memorial Highway"

Commemorates six African-American Army units created in 1866 (9th and 10th Cavalry, 38th, 39th, 40th, and 41st Infantry Regiments), later reorganized in 1868 as the 24th and 25th Infantry Regiments

Recognizes Buffalo Soldiers stationed at Fort Lyon and Fort Garland, Colorado who protected settlers, built infrastructure, and earned 23 Medals of Honor and 40 Certificates of Merit with the Army's lowest desertion rate

Authorizes the Colorado Department of Transportation to accept gifts, grants, and donations for placement of signage marking the memorial highway

Authorizes CDOT to explore a cooperative agreement with El Paso County Commissioners for maintenance of the memorial highway markings
