Establishes the Transportation Infrastructure Investment Fund to provide economic assistance for road and transportation infrastructure improvements that attract new businesses or expand existing businesses and create significant permanent full-time jobs.

Requires funded improvements to occur within public right-of-way or on public land, directly benefit the traveling public, have at least one public endorser, meet employment standards, be financially stable, and serve a public purpose.

Creates a 9-member Transportation Infrastructure Investment Fund Council appointed by the President Pro Tempore of the Senate, Speaker of the House, and Governor (with geographic and political party balance requirements) to review applications and recommend awards.

Requires the Secretary of the Department of Transportation and Secretary of State to authorize fund expenditures only after determining that businesses and improvements meet eligibility criteria and comply with departmental regulations.

Takes effect upon enactment and becomes operational 10 days after the Department of Transportation publishes notice that final implementing regulations have been promulgated.
