Submits a constitutional amendment to Missouri voters at the general election in November 2010 (or a special election called by the governor) to repeal and replace Section 29 of Article IV of the Missouri Constitution.

Replaces the Highways and Transportation Commission with a single Director of Transportation appointed by the governor with advice and consent of the state senate.

Removes requirements that the commission have a specified number of members, qualifications, compensation, and terms, and the prohibition against more than one-half of members being from the same political party.

Maintains the department's authority over the state highway system, aviation, railroads, mass transportation, ports, waterborne commerce, and authority to limit access to transportation facilities for public safety.

Specifies that current Highways and Transportation Commission members shall continue serving until a director is appointed and confirmed by the senate.
