Adds "sexual orientation" as a protected class in Missouri's human rights law, defined as male or female heterosexuality, homosexuality, or bisexuality by inclination, practice, identity, or expression, or having a self-image or identity not traditionally associated with one's gender.

Prohibits discrimination based on sexual orientation in employment, housing, public accommodations, and financial services for mortgages and loans.

Applies protections to the Missouri Commission on Human Rights, giving it authority to investigate complaints and enforce civil rights protections related to sexual orientation discrimination.

Establishes a human rights fund to receive civil penalties and offset expenses related to compliance with federal regulations, particularly Department of Housing and Urban Development requirements.

Updates rulemaking procedures to comply with Missouri's Administrative Procedure Act (Chapter 536), making rules subject to general assembly review and potentially void if the rulemaking authority is found unconstitutional after August 28, 2011.
