Designates a portion of Interstate 70 from the western city limits of a city not within a county to mile marker 248 as the "Mark Twain Highway".

Designates the Missouri portion of the Poplar Street Bridge crossing the Mississippi River on Interstate 55, 64, 70, and U.S. Route 40 as the "Congressman William L. Clay, Sr. Bridge".

Designates the new Interstate 70 bridge crossing the Mississippi River between St. Louis and southwestern Illinois as the "Stan Musial Memorial Bridge".

Creates multiple additional highway and bridge designations throughout Missouri, including memorials for fallen soldiers and public servants, with all signage costs to be paid through private donations.

Requires the Department of Transportation to erect and maintain signs for all designated highways and bridges.
