Creates the Mississippi Smokefree Air Act of 2016, establishing comprehensive prohibitions on smoking in enclosed public places, places of employment, private clubs, and certain outdoor areas effective July 1, 2016.

Prohibits smoking in all enclosed public places including bars, restaurants, health care facilities, educational facilities, gaming facilities, and retail establishments; prohibits smoking in all enclosed workplaces without exception.

Prohibits smoking outdoors within 20 feet of entrances and windows of smokefree buildings, in outdoor restaurant and bar seating areas, around playgrounds and public transportation facilities, and in outdoor workplaces where two or more employees are present.

Establishes penalties including fines up to $50 for individuals smoking in prohibited areas and fines of $100-$500 for owners/operators of non-compliant establishments, with possible suspension or revocation of business licenses; violations declared a public nuisance subject to injunctive relief.

Requires posting of "No Smoking" signs at entrances and throughout affected areas, removal of ashtrays from prohibited areas, and prohibits retaliation against employees or customers for exercising smokefree air rights; repeals the previous Mississippi Clean Indoor Air Act.
