Designates a segment of U.S. Highway 49/49E/49W from Jackson's corporate limits at Medgar Evers Boulevard in Hinds County northerly to U.S. Highway 61 in Clarksdale, Coahoma County, as an historic memorial blues highway, spanning nine counties (Hinds, Madison, Yazoo, Holmes, Leflore, Humphreys, Sunflower, Tallahatchie, and Coahoma).

Requires municipalities and counties along the route to work with the Mississippi Blues Commission to designate historical sites and events along the highway to create a structured tour for tourists and targeted groups.

Mandates the Mississippi Department of Transportation maintain the designated highway segment to standards appropriate for a historic highway.

Authorizes MDOT to enter interlocal agreements with all counties encompassing the route to support upkeep and maintenance of the historic memorial blues highway.

Takes effect July 1, 2023.
